Recombinants then grow on ampicillin but not on tetracycline. This is <strong>insertional inactivation<\/strong>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">With the <em>lacZ<\/em> gene as a marker, recombinant colonies are <strong>white<\/strong> and non-recombinants are <strong>blue<\/strong>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Other cloning vectors and methods to remember:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><em>Agrobacterium tumefaciens<\/em> delivers T-DNA into dicot plants through its Ti plasmid.<\/li>\n\n\n\n<li>Retroviruses are used as vectors for animal cells.<\/li>\n\n\n\n<li>Bacterial cells are made competent with calcium ions and heat shock.<\/li>\n\n\n\n<li>Microinjection is used for animal cells; the gene gun (biolistics) is used for plant cells.<\/li>\n<\/ul>\n\n\n\n<h2 id=\"pcr-steps-and-gel-electrophoresis\" class=\"wp-block-heading\">PCR Steps and Gel Electrophoresis<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">The three PCR steps are:<\/p>\n\n\n\n<ol class=\"wp-block-list\">\n<li><strong>Denaturation<\/strong> \u2013 the two DNA strands separate at high temperature.<\/li>\n\n\n\n<li><strong>Annealing<\/strong> \u2013 primers bind to the template.<\/li>\n\n\n\n<li><strong>Extension<\/strong> \u2013 <em>Taq<\/em> polymerase, from <em>Thermus aquaticus<\/em>, adds nucleotides.<\/li>\n<\/ol>\n\n\n\n<p class=\"wp-block-paragraph\">In gel electrophoresis, DNA is negatively charged and moves towards the <strong>anode<\/strong>. Smaller fragments travel farther through the agarose gel. Bands are seen as bright orange under UV light after staining with ethidium bromide. Cutting the band out and extracting the DNA is called elution.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">To isolate DNA, the cell wall is broken with lysozyme (bacteria), cellulase (plants) or chitinase (fungi). The DNA is finally precipitated with chilled ethanol.<\/p>\n\n\n\n<h2 id=\"applications-the-facts-that-repeat\" class=\"wp-block-heading\">Applications: The Facts That Repeat<\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Bt cotton:<\/strong> the <em>cry<\/em> genes come from <em>Bacillus thuringiensis<\/em>. <em>cryIAc<\/em> and <em>cryIIAb<\/em> control cotton bollworms; <em>cryIAb<\/em> controls the corn borer. The inactive protoxin is activated by the alkaline pH of the insect gut.<\/li>\n\n\n\n<li><strong>RNA interference<\/strong> protects tobacco from the nematode <em>Meloidogyne incognita<\/em>.<\/li>\n\n\n\n<li><strong>Human insulin:<\/strong> Eli Lilly produced chains A and B separately in <em>E. coli<\/em> in 1983 and joined them by disulphide bonds. Mature insulin lacks the C-peptide.<\/li>\n\n\n\n<li><strong>Gene therapy:<\/strong> first given in 1990 to a four-year-old girl with ADA deficiency.<\/li>\n\n\n\n<li><strong>Molecular diagnosis:<\/strong> PCR and ELISA detect infections early.<\/li>\n\n\n\n<li><strong>Rosie<\/strong>, the first transgenic cow (1997), produced milk enriched with human \u03b1-lactalbumin.<\/li>\n\n\n\n<li><strong>GEAC<\/strong> decides on the validity of GM research and the safety of GM organisms.<\/li>\n\n\n\n<li><strong>Biopiracy:<\/strong> the Basmati rice patent case of 1997 is the standard example.<\/li>\n<\/ul>\n\n\n\n<h2 id=\"worked-pyq-pattern-questions\" class=\"wp-block-heading\">Worked PYQ-Pattern Questions<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Q1.
